Biotherapeutics works on projects from five Therapeutic Areas (TA) and enables discovery of new therapeutic concepts in collaboration with TA discovery counterparts. Supporting the CAR-T Platform group. This position will be accountable for early discovery efforts to engineer CAR-T cells for initial functional characterization in conjunction with our team and Oncology Therapeutic Area stakeholders. Key responsibilities include, but are not limited to: daily maintenance of cell cultures to enable the production of lentiviral particles and/or CAR-T cells; working knowledge of viral-based production technologies and quality control (QC) techniques; ability to transduce primary human T-cells with both lenti- and adeno-associated viral particles; utilize flow cytometric techniques to determine transduction efficiency and to perform related QC measurements of engineered cells; provide functional and intellectual support to the CAR-T cell group within Biotherapeutics.
Essential Functions:
  • Strong experience in research-scale lentivirus vector/particle generation
  • Working knowledge of assays that measure infectious and physical viral titer
  • Expertise in primary T cell gene transfer via lentiviral transduction
  • Isolation of primary human immune cells, including T-lymphocytes, from human blood and subsequent aseptic cell culture
  • Phenotypically characterize genetically modified primary T-lymphocytes using flow cytometric instrumentation
  • Viral vector-based or electroporation-based modification of tumor cell lines and primary human T-cells
  • Maintain an electronic lab notebook and contribute to support documentation in line with company standards
  • Support the CAR-T R&D; team in JBIO to enable timely development and delivery of novel molecular entities to therapeutic area stakeholders
  • Self-motivated and eager to take on temporary responsibilities outside of initial job description
